
Highly Sensitive LabelingReagents for Scarce Natural Products
Scarce natural products that possess unique biological activities have been ideal drug leads for decades. However, their identification and structural determinations are problematic owing to sample amount limitation. Inspired by an extremely rare natural product yaku’amide B (<b>10</b>), highly sensitive labeling reagents that would be powerful tools for scarce natural product chemistry were designed and synthesized in this study.
By fusion with the key structural motif for the structural revision of <b>10</b>, the detection sensitivities of amino acid labeling reagents were drastically enhanced in LC-MS analysis. These advanced labeling reagents enabled the detection of infinitesimal amounts of amino acids and peptide hydrolysates. This sensitivity-enhancement design concept was also applicable to reagents for labeling saccharides and reactivity-guided isolation of electrophilic natural products. Details of these reagents, including their practical preparations and extended applications, are also provided.

Data-driven adaptive modeling method for industrial processes and its application in flotation reagent control
In real industrial processes, new process “excitation” patterns that largely deviate from previously collected training data will appear due to disturbances caused by process inputs. To reduce model mismatch, it is important for a data-driven process model to adapt to new process “excitation” patterns. Although efforts have been devoted to developing adaptive process models to deal with this problem, few studies have attempted to develop an adaptive process model that can incrementally learn new process “excitation” patterns without performance degradation on old patterns.
In this study, efforts are devoted to enabling data-driven process models with incremental learning ability. First, a novel incremental learning method is proposed for process model updating. Second, an adaptive neural network process model is developed based on the novel incremental learning method. Third, a nonlinear model predictive control based on the adaptive process model is implemented and applied for flotation reagent control. Experiments based on historical data provide evidence that the newly developed adaptive process model can accommodate new process “excitation” patterns and preserve its performance on old patterns. Furthermore, industry experiments carried out in a real-world lead-zinc froth flotation plant provide industrial evidence and show that the newly designed controller is promising for practical flotation reagent control.
Photoswitchable affinity reagents: Computational design and efficient red-light switching
Photo-controlled affinity reagents seek to provide modular spatiotemporal control of bioactivity by conferring photo-switchability of function on an affinity reagent scaffold. Here we used Rosetta-based computational methods to screen for sites on the Fynomer affinity reagent structure for attachment of photoswitchable cross-linkers. Both established UV-based cross-linkers (azobenzene-iodoacetamide (IAC)) and an azonium-based efficient red light switchable cross-linker, piperazino-tetra-ortho-methoxyazobenzene (PIP), were then tested experimentally.
Several sites compatible with Fynomer function were identified, including sites showing rapid (<10s) red light (633 nm) modulation of function. While a range of overall target binding affinities were observed, the degree of photo-switchability of Fynomer function was generally small (<2-fold). Computational models suggest that local flexibility limits the degree of switching seen in these designs.
The tea-bag protocol for comparison of Fmoc removal reagents in solid-phase peptide synthesis
Several factors have influenced the increasing presence of peptides as an important class of Active Pharmaceutical Ingredients. One is the continued development of synthetic methodologies for peptide synthesis. Herein, we investigated the Fmoc removal step, using the tea-bag strategy.
In this regard, three different secondary amines: piperidine, 4-methylpiperidine, and piperazine, were evaluated. As a result of this study, 4-methyl piperidine showed to be an excellent alternative to the usually used piperidine in terms of purity and compliance with green chemistry principles as well.
Photochemical Functionalization of Heterocycles with EBX Reagents; C-H Alkynylation versus Deconstructive Ring Cleavage
The development of novel methodologies for the functionalization of saturated heterocycles is highly desirable. Herein, we report a cheap and efficient photochemical method for the C-H functionalization of saturated O -heterocycles, as well as the deconstructive ring-cleavage of S -heterocycles, employing hypervalent iodine alkynylation reagents (Ethynylbenziodoxolones, EBX).
This photochemical alkynylation is performed utilizing phenylglyoxylic acid as the photoinitiator, leading to the corresponding products in good to high yields, under household fluorescent light bulb irradiation. When O -heterocycles were employed, the expected α-C-H alkynylation took place. In contrast, oxidative ring-opening to form a thioalkyne and an aldehyde was observed with S -heterocycles. Preliminary mechanistic experiments are presented to give first insights into this puzzling divergent reactivity.
